NEWARK, N .J. [A.P.] — Got a cold? Dr. John T. Connell will pay you $30 to $50 a day to sit in his office and sneeze. Connell, a specialist in nose problems and allergies, is one of dozens of experts throughout the country who test cold remedies for drug companies trying to find out how well their products work. “ I’m an independent investi gator,” he said. “ They come to me. 1 design an experiment to try to solve their problems and then we talk about a fee .” Connell ran an ad, “ Head Colds W anted,” on Jan. 9 in connection with his latest test. Since then, he has heard from more than 600 suffering volunteers, of whom 200 qualified. “ I don’t want people who are only doing it for the money,” he said in a telephone interview from his Englewood office. “ They have to be between 18 and 60, and not have a history of heart disease, high blood pressure or other comp lications." On a recent wintry day. Connell ushered eight cold victims into his office at 8:20 a.m . and fitted their nostrils with plastic cups connected to a machine that m easures nasal airflow. BIOLOGICAL, CULTURAL, GENETIC, AND CHEMICAL CONTROL OF PIERCE’S DISEASE: PRODUCTION AND SCREENING OF XYLELLA FASTIDIOSA TRANSPOSON PATHOGENICITY AND ATTACHMENT MUTANTS Project Leaders:

INTRODUCTION One of our projects involved the development of a transformation and transposon mutagenesis systems for the bacterium that causes Pierce's disease (PD), Xylella fastidiosa (Xf). We now have a random transposon mutagenesis system working for Xf (Guilhabert, et al. 2001) and recently we have developed an E. coli/Xf plasmid shuttle vector. We are currently assessing the stability of t...

Application of bacterial shuttle vectors in designing new vaccines against infectious diseases: brief report

Background: Today, several vaccines have been developed to prevent infectious diseases. The older first-generation vaccines may have many problems. In this regard, genetic engineering plays an important role using tools such as shuttle vectors to develop recombinant DNA vaccines that usually include plasmid constructed so that can propagate in two different host species. Yeast expression vectors using RNA polymerase III promoters.

A series of Saccharomyces cerevisiae--Escherichia coli shuttle vectors is described in which small RNAs can be stably expressed in yeast from two different promoters for RNA polymerase III transcription. The vectors are available in either high- or low-copy-number forms with either URA3, HIS3, or TRP1 selection markers, and are based on a previously described set of plasmid vectors [Sikorski an...
